Semaglutide Dosage Guide: How Much to Take

Complete dosage information for Semaglutide including starting dose, maintenance dose, titration schedule, and administration guidance.

Dosage at a Glance

Starting Dose
0.25
mg
Typical Dose
1.7-2.4
mg
Max Studied Dose
2.4
mg
Frequency: Once weekly (injectable); once daily (Rybelsus oral)

Dosing Notes

For Wegovy: Start at 0.25mg weekly, titrate by 0.25mg every 4 weeks up to 2.4mg maintenance dose. For Ozempic: Max dose is 2mg. Oral Rybelsus dose is 3mg daily titrating to 14mg.

Titration Schedule

Gradual titration reduces GI side effects. Follow this schedule, adjusting based on tolerability.

WeekDoseNotes
Week 10.25mgStarting dose — assess tolerability
Week 50.5mgIncrease if tolerated
Week 91mgIncrease if tolerated
Week 131.7mgIncrease if tolerated
Week 172.4mgMaintenance dose

Administration Routes

  • Subcutaneous injection (abdomen, thigh, upper arm)
  • Oral tablet (Rybelsus)

What to Do If You Miss a Dose

General guidance: If you miss a dose, take it as soon as you remember — unless it's almost time for your next dose. Never double up. For weekly peptides, a 1–2 day delay is generally acceptable without restarting titration.

How to Reconstitute Semaglutide

If you have lyophilized (freeze-dried) Semaglutide powder, it must be reconstituted with bacteriostatic water (BAC water) before use. Here is the standard reconstitution process:

1
Gather supplies
Lyophilized peptide vial, bacteriostatic water (BAC water), insulin syringe, alcohol swabs
2
Clean surfaces
Wipe the rubber stoppers of both vials with alcohol swabs and allow to air dry
3
Draw BAC water
Draw the desired volume of BAC water into an insulin syringe
4
Add BAC water slowly
Insert needle into the peptide vial at an angle; let BAC water run down the inside wall — do NOT inject directly onto the powder
5
Swirl gently
Gently swirl (do NOT shake) until the powder fully dissolves — the solution should be clear
6
Store correctly
Refrigerate immediately at 2–8°C (36–46°F). Use within 28 days.

BAC Water Volume Reference

The amount of BAC water you add determines the concentration per unit (IU or tick) on your syringe. Common configurations:

Vial SizeBAC Water AddedConcentrationVolume per mg
5mg1mL5mg/mL0.20mL per 1mg
5mg2mL2.5mg/mL0.40mL per 1mg
10mg1mL10mg/mL0.10mL per 1mg
10mg2mL5mg/mL0.20mL per 1mg

Using a 100-unit (1mL) insulin syringe: each 10 tick marks = 0.1mL. Adjust your draw based on the concentration above.

Storage & Shelf Life

Lyophilized (dry powder)
Temperature: 2–8°C refrigerated
Shelf life: 12–24 months
Can withstand room temp for shipping (few days)
Reconstituted (liquid)
Temperature: 2–8°C refrigerated only
Shelf life: 28 days
Never freeze reconstituted peptide — degrades quickly
